Diversity and ¢eld site variation of indigenous populations of soybean bradyrhizobia in Japan by ¢ngerprints with repeated sequences RSK and RSL

چکیده
Two hundred and thirteen isolates of soybean bradyrhizobia indigenous to six field sites in Japan were characterized using hybridization probes RSK, RSL, nifDK and hupSL from Bradyrhizobium japonicum and indole-3-acetic acid production to clarify diversity and endemism of their population structures. Significant diversities and site-dependent variations were observed in terms of Bradyrhizobium species, hup genotype and fingerprints with repeated sequences RSK and RSL. The fingerprints at one site with no history of soybean cultivation were less diverse than those at other sites. Even within the populations of B. japonicum hup3 isolates, which were commonly indigenous to the six field sites, several RSK copies clustering around nif genes were highly conserved. The results suggest that soybean bradyrhizobia may be diversified in individual fields as associated with host plants and local soil conditions. z 1999 Federation of European Microbiological Societies. Soybean (Glycine max [L.] Merr.) establishes a symbiotic relationship by infection with soybeannodulating bacteria and subsequent root nodule formation. Soybean acquires atmospheric nitrogen as ammonia through the symbiotic nitrogen fixation by soybean-nodulating rhizobia in the root nodules. The inoculation of soybean with bradyrhizobia that has high ability of nitrogen fixation is considered ...

Mitochondrial DNA (mtDNA) has been used extensively to study population genetics because it has the unique features of maternal inheritance, a relatively fast rate of evolution and lack of recombination. A total of 82 unrelated sheep from 10 Iranian indigenous sheep breeds were investigated to determinate the maternal genetic diversity using a sequence of a 685 bp segment of the displacement lo...
